When a host on the network doesn't know the MAC address of the device it wants to reach, it will broadcast an ARP request. All hosts on the segment will receive this ARP request and one of them will respond with an ARP reply. In this ARP reply, you will find the IP address and MAC address of the destination host. Once the host that sent the ARP requests receives this information, it will be able to reach the destination.
